Tungsten and Molybdenum Compound Application in Anti-cancer Drug

Molybdenum is a trace element of life required by living organisms. Molybdenum deficiency in the human body can cause a series of diseases, such as Keshan disease, kidney stones, Kashin-Beck disease, high blood pressure and diabetes. In particular, the lack of molybdenum can lead to an increase in the incidence of cancer.

According to the literature, in the high-incidence areas of cancer such as esophageal cancer, digestive tract cancer, stomach cancer, liver cancer, rectal cancer, lung cancer and breast cancer, the content of molybdenum in serum, urine samples and hair is significantly lower than that in low-incidence areas. The incidence of anterior gastric cancer in molybdenum-deficient animals was significantly higher in normal animals than in normal animals after receiving a certain dose of nitroso compounds.

In living organisms, metal molybdenum is an antagonistic element of metallic copper, which has long been accepted. Since 1991, research on anticancer and anti-tumor angiogenesis of ammonium tetrathiomolybdate has begun. The growth and metastasis of malignant tumors are closely related to the blood vessels in the tumor area. When the tumor grows to about 2 mm, new blood vessels are needed to provide the oxygen and nutrients needed for its growth, and copper is the formation of new blood vessels. The nutrients necessary for growth, that is, the formation of new blood vessels depends on copper. If the content of copper ions in the tumor area is reduced, the formation and growth of blood vessels in the region can be prevented. Therefore, treatment of tumors with neovascularization as a target has become a very active research in recent years.

At present, the use of ammonium tetrathiomolybdate as a target for neovascularization has entered the second phase of clinical, and achieved a good anti-cancer and anti-tumor effect, prolonging the life expectancy of patients.

However, ammonium tetrathiomolybdate is very unstable and extremely sensitive to light, water and oxygen. Therefore, it needs to be placed in an anhydrous dry, dark, oxygen-free environment, generally stored in an inert argon. Within 8 weeks, the ammonium tetrathiomolybdate in the capsule retained only 90% of the drug's effectiveness. Therefore, it is imperative to seek new anti-cancer and anti-tumor drugs that are highly effective, low-toxic, superior or specific to current clinical drugs, and have certain stability.

Tungsten is a homologous element of molybdenum and is also the active center of tungsten oxygen transferase. In many organisms, tungstotransferase coexists with molybdenum oxytransferase and has similar biochemical functions. Therefore, tungsten and molybdenum compound can be used to prepare highly effective anticancer and antitumor drugs.

It has been verified by scientific researchers that the tungsten and molybdenum compound has good water solubility and fat solubility. In the scientific experiment, 10 Kunming mice were taken as a group, and the tumor cells were taken from the abdominal cavity of the mice. After treatment, the cells were diluted to 5×10 6 /ml, under sterile conditions, 0.2 ml / inoculation in the left forelimb of the mouse, 24 hours after inoculation, group administration, intraperitoneal injection once a day for 10 consecutive days. On the 12th day, the tumor was weighed and the average tumor weight was calculated for each group. The standard deviation was P value. Cyclophosphamide was used as a positive control group. LuI 100mg/kg, 50mg/kg and positive control inhibited mouse sarcoma S180, and the inhibition rates were 68.7 (P<0.01), 32.2 (P<0.01), and 64.5 (0.01), respectively. Experiments show that the tungsten and molybdenum compound has an excellent inhibitory effect on mouse sarcoma S180, and it is promising for clinical targeted drugs in the future.
